ABSTRACT

Diffquick staining is a rapid staining method, usually used for histological staining to differentiate
smears and from fine needle aspirates. One of the solutions for diffquick staining is eosin. Eosin has
the function of coloring the cytoplasm. Eosin has toxic properties so natural dyes are needed that
have eosin-like properties. Shoe flower (Hibiscus rosa-sinensis L.) is often used as a natural dye which
contains anthocyanins. The aim of this research was to determine the effect of storage treatment on
hibiscus extract in staining the cytoplasm of buccal smear epithelial cells using the diffquick staining
method. The research location was carried out at the STIKes Mitra Keluarga cytohistotechnology
laboratory on 1 February-25 Juny 2024 with the research design used being experimental. The
sample used buccal smears from 15 students with a ttal sample of 30. The sampling technique used
purposive sampling and the production of hibiscus flower extract using the maceration method. The
results of the research showed that the storage treatment at a 25°C temperature obtained low results,
namely an average value of 2.6, while the results were close to the control solution, namely at a
temperature of 7°C. namely an average value of 2.7. Hibiscus flower extract can color well for 3 days
at a temperature of showcase temperatureon buccal smear preparations using the diffquick staining
method.
